Mechanism

NGF (Nerve growth factor)

The first neurotrophin discovered (Rita Levi-Montalcini, Nobel Prize 1986). Essential for the survival and growth of sympathetic and sensory neurons. In the adult brain it sustains cholinergic neurons of the basal forebrain, whose degeneration is an early hallmark of Alzheimer's. Target of experimental neurotrophin therapy.

Alzheimer's and neurodegeneration

protects Moderate phase II RCT (AA)
Experimental target

Trials with gene therapy introducing NGF into the nucleus basalis of Meynert show preservation of cholinergic neurons and stabilization of cognitive decline in moderate Alzheimer's. Preliminary data.

Peripheral nerve regeneration

promotes Moderate preclinical (A)
Peripheral regeneration

NGF accelerates peripheral nerve regeneration after injury in animal models. Investigated in diabetic neuropathy and in traumatic injuries.

Lion's mane (Hericium erinaceus)

increases (possible) Low-moderate preclinical + pilot RCT (A)
Nutraceutical pathway

Lion's mane extracts induce NGF synthesis in cell cultures. A pilot RCT in mild cognitive impairment showed improved cognitive scores; preliminary data.

Falling in love

increases (transient) Low observational (A)
Basis of falling in love

Studies by Enzo Emanuele show a transient elevation of plasma NGF in recent lovers (first 6 months), which then returns to baseline levels. Biochemical correlate of falling in love.
